Structure
Berger et al. started with the gene of the membrane bound human placental alkaline phosphatase (PLAP). The secreted form (SEAP) was derived by insertion of a translational terminator (stop codon) after aa 489 using an oligonucleotide-directed mutagenesis procedure. This procedure generated a unique Hpa I site. A 2.1 kb Hind III/EcoR I fragment containing the SEAP gene was transferred from pBC/PL/SEAP (Berger et al. 1988) to pBluescript IIKS+. A 2.1 kb Hind III / Xba I fragment was then transferred into the pRc/CMV plasmid to form the pCMV/SEAP plasmid vector (Total 7582 bases).
