Mode of Growth: loosely adherent (usually some cells are found floating). The growth rate seems to improve with increased density of plating. About 8 x 105 cells in a T25 take 3-5 days to reach confluency. KY cells grow IL-2 dependent.

Method of Passage: nonenzymatic dissociation (PBS + 2% EDTA) to detach the cells from the surface

Culture Medium: R10 (= RPMI 1640 with 2 mM L-glutamine (300 µg/ml), 10% FBS, 1% pen/strep (100 U/ml penicillin, 100 µg/ml streptomycin), 0.1 mM 2-mercaptoethanol (7µl 2-ME per liter media). Add IL-2 stock solution (105 U/ml) directly to cells. When first growing KY clones (e.g. when reviving frozen cells), start with 103 U/ml, passing a few times before gradually weaning down to 50 U/ml, which is adequate for normal maintenance.

Freeze Medium: culture medium + 10% DMSO
