Station 5: Prepare Slides
Fixed call suspensions are dropped onto slides in a temperature
and humidity controlled chamber. As the cells dry, the membrane
flattens and the chromosomes spread out. Consistent dry conditions
are necessary for quality chromosome morphology. Slides are "aged"
at 60 degrees C for at least 12 hours increase the quality and
definition of the banding.
Although the procedures are methodical, the skill of the technologist
is important to the quality of specimen preparation. Troubleshooting
the variables within the process is an invaluable skill of an
experienced technician.
