Once the cells have swollen, a fixative is added to preserve the cells in the swollen state. This fixative also stabilizes the cell membrane and the chromatin. This is critical in preparing the chromosomes for the banding process. Several changes of fix are needed to remove excess water and debris from harvested cells.
